Technologies Imperacer | The Immuno-PCR Platform
Imperacer® is an ultra-sensitive immunoassay platform developed by chimera biotec. It combines the specificity of enzyme-linked immunosorbent assays (ELISA) with the amplification power of polymerase chain reaction (PCR), a method known as Immuno-PCR. This combination allows for the detection of analytes at concentrations significantly lower than those detectable by conventional ELISA, achieving sensitivities in the sub-picogram to femtogram per milliliter range.
Imperacer® has been applied in various fields, including the quantification of cytokines, detection of biogenic amines, and measurement of therapeutic antibodies in pharmacokinetic studies. Its high sensitivity and robustness make it a valuable tool for both research and clinical applications.

Imperacer® Technology
The Imperacer® technology utilizes antibody-DNA conjugates for target detection, followed by exponential DNA amplification to generate the signal. This approach not only enhances sensitivity but also provides a broad dynamic range, enabling the quantification of analytes over several orders of magnitude. Additionally, the platform is designed to be robust across various sample matrices (CSF, saliva) and requires minimal sample volumes, making it suitable for applications where sample availability is limited.

Ultra-Sensitive Immunoassay Platform with Exponential Signal Amplification
Leveraging Imperacer® Technology for Minimal Sample Volume and High Sensitivity in Immunoassays
Our proprietary immunoassay platform Imperacer® combines the preeminent specificity of antibody-antigen binding with the extremely powerful signal amplification of DNA analytics, using our patented antibody-DNA conjugates.
A double-stranded DNA-tag allows for exponential signal amplification of immunoassay binding events by PCR. This exponential amplification results in ultra sensitive assays with very broad dynamic ranges and – in consequence – to very dilution tolerant assays.
Imperacer® allows modification of the most sensitive assay procedure toward an assay with the minimal sample volume requirement at the needed level of sensitivity by customized AnySource® sample dilution.
Sensitivity can be employed for immunoassay quantification from microliter (µl) volumes of neat sample, still maintaining the needed sensitivity requirement of the study in trials involving microsampling or rare matrix samples.

Ultra sensitive in 5 steps of Imperacer
Streamlined immunoassay workflow with enhanced sensitivity and minimal background using AnySource® technology
Step 1: Microplates are coated with a capture antibody specific to the target analyte.
Step 2: A blocking solution is applied to reduce non-specific binding and background noise.
Step 3: Samples containing the analyte are added, allowing the target molecules to bind to the capture antibodies.
Step 4: Proprietary antibody-DNA conjugates are introduced, binding to the captured analyte and forming a complex.
Step 5: Real-time quantitative PCR (qPCR) is performed to exponentially amplify the DNA marker, resulting in a highly sensitive detection signal.
Chimera´s proprietary AnySource® technology allows high sample dilution to reduce background to a minimum – but still detecting ultra sensitively with our DNA-antibody conjugates.
Our advanced immunoassay workflow is designed to maximize sensitivity while minimizing background interference. Starting with the coating of 96-well plates with capture antibodies, the process ensures specific binding of analytes. The use of a blocking solution further reduces non-specific background signals.
Upon adding samples, analytes bind to the capture antibodies, forming the initial complex. This is followed by the introduction of Chimera’s proprietary DNA-antibody detection conjugates, completing the binding complex. The final step involves real-time qPCR, which exponentially amplifies the signal intensity, ensuring ultra-sensitive detection.
Chimera’s AnySource® technology allows for high sample dilution, effectively reducing background noise while maintaining the sensitivity required for accurate detection. This streamlined workflow ensures reliable and precise results, even with minimal sample volumes.
